Epidemiological and molecular characterization of Enterobius vermicularis cases in Mersin, Turkey: An analysis based on mitochondrial DNA cox1 gene sequences

Enterobius vermicularis is among the most common intestinal helminths, especially in children. Mitochondrial cox1 gene analyses revealed three genotypes (A, B, C). We aimed to evaluate the epidemiological characteristics and cox1 genotypes of E. vermicularis cases diagnosed at Mersin University Hospital. Between January 2017 and December 2023, 1496 samples were examined using the cellophane tape method, and 75 positives were identified. Epidemiological data were retrospectively analyzed. DNA was isolated from 30 positive samples via the phenol–chloroform method, and nested PCR products of the cox1 gene were sequenced. Phylogenetic analyses were performed using GenBank sequences. In a sample of patients with gastrointestinal signs, the positivity rate for E. vermicularis was 5 % (75/1496). Among the positive cases, 93.3 % were pediatric, with a mean age of 8.7 ± 7.7 years, and the highest prevalence (9 %) was observed in the 5–10 age group. The most commonly reported symptoms included abdominal pain (49.3 %), perianal itching (40 %), and nocturnal drooling (33.3 %). Alpha Human Papillomavirus Types and Genotype Distribution in Psoriasis Patients: A Cross-Sectional Regional Study in Turkey.

Background: Human papillomaviruses (HPVs) are members of the P apillomaviridae family, characterized by circular, double- stranded DNA. T hey exhibit strict tropism, infecting either mucosa or skin. T he role of HPV in the etiology of psoriasis remains unclear. Objectives: T he aim of this study was to determine the presence of alpha human papillomavirus in the skin scrapings of patients with psoriasis and its genotype distributions. Methods: T he study employed a cross-sectional design and included 53 psoriasis patients and 47 healthy controls from December 2020 to May 2021. After DNA isolation from skin scrapings, the L1 gene region of HPV was amplified using MY09/11 and GP5 +/6 + consensus primer sequences. T he HPV genotypes were determined through direct DNA sequence analysis of polymerase chain reaction (PCR) products. Results: HPV DNA was detected in 34% of psoriasis patients and 29.8% of healthy individuals. Among the patients, HPV18 was found in 11.1%, HPV31 in 38.9%, and HPV81 in 50%. Evaluation of cutaneous leishmaniasis cases in Mersin, Türkiye: Impact of migration and the COVID-19 pandemic

Introduction: Cutaneous leishmaniasis (CL) is a significant public health concern in Türkiye, especially in the Southeastern Anatolia and Mediterranean regions. This study evaluated the epidemiological characteristics of CL cases diagnosed at Mersin University Hospital, including the impact of migration and the COVID-19 pandemic. Methodology: The diagnosis of CL was confirmed by microscopic examination of lesion smears. In this study, data from 144 patients diagnosed with CL between January 2011 and December 2022 were evaluated retrospectively. Demographic characteristics of the patients, temporal distribution (by month and year), and lesion features (location, number, and duration) were included. Results: CL was confirmed in 32.9% of patients (144/438). Of the 144 patients, 82 (56.9%) were Turkish citizens and 62 (43.1%) were Syrian refugees. The mean age was 20.7 ± 19.8 (1-78) years. CL was detected more frequently in females (52.1%, 75/144) and in the 0–20-year age group (64.6%, 93/144). Effects of Sequential Antimicrobial Phases on Root Canal Microbiome Dynamics in Two-Visit Treatment of Primary Apical Periodontitis: A Longitudinal Experimental Study.

Abstract: Background: Effective management of primary apical periodontitis depends on understand- ing the dynamic interactions within the root canal microbiome. This study aimed to investigate the effect of sequential antimicrobial phases on the root canal microbiome during a two-visit treatment approach, with a focus on calcium hydroxide medication. Methods: Samples were collected from three teeth across four treatment phases: initial infection (S1), after chemomechanical preparation (S2), after intracanal medication (S3), and after a final flush (S4). DNA was extracted, and the V3–V4 regions of the 16S rRNA gene were sequenced using Illumina MiSeq. Sequencing data were analyzed with QIIME 2, and differentially abundant taxa were identified using linear discriminant analysis effect size (LEfSe). Results: While microbial community composition did not differ significantly between phases, the Firmicutes/Bacteroidetes ratio decreased after the antimicrobial stages. LEfSe analysis revealed higher abundances of Lactobacillales, Arthrobacter, and Veillonella in the untreated (CMP) group. Pneumocystis jirovecii İzolatlarının Dihidropteroat Sentaz (DHPS) Gen Mutasyonları ve Mitokondriyal Büyük Alt Birim (mtLSU) rRNA Genotip Varyasyonlarının Belirlenmesi

Pneumocystis jirovecii, HIV/AIDS ve immün sistemi baskılanmış hastalarda Pneumocystis pnömonisine (PCP) neden olan atipik bir fungustur. PCP profilaksi ve tedavisinde sülfa ve sülfon grubu içeren ilaçlar yaygın olarak kullanılır. Özellikle trimetoprim sulfametoksazol (TMP-SMX)’un, uzun süreli kullanılması P.jirovecii dihidropteroat sentaz (DHPS) geninde ilaç direnci ile ilişkili belirli nokta mutasyonlara neden olmaktadır. Ayrıca, DHPS ve mitokondriyal büyük alt birim [mitochondrial large subunit (mtLSU)] rRNA genotip karakterizasyonu P.jirovecii epidemiyolojisi hakkında önemli veri sağlamaktadır. Bu çalışmada, Mersin Üniversitesi Hastanesinde, immün sistemi baskılanmış hastalardan izole edilen P.jirovecii izolatlarının DHPS ve mtLSU rRNA gen bölgesi polimorfizmlerinin araştırılması amaçlanmıştır. Bu çalışmaya, Ağustos 2016-Şubat 2018 tarihleri arasında 96 hasta örneğinden mtLSU-rRNA nested PCR ile pozitif bulunan 16 P.jirovecii örneği dahil edilmiştir. P.jirovecii mtLSU rRNA genotipleri dizi analizi yapılarak 85. ve 248. nükleotit pozisyonundaki polimorfizmlere göre belirlenmiştir. Genetic diversity and antimicrobial susceptibility of motile aeromonads isolated from rainbow trout (Oncorhynchus mykiss, Walbaum) farms

A total of 22 motile Aeromonas strains were detected in 48 (18.53%) of 259 fish and 6 (10.71%) of 56 water samples obtained from seven commercial rainbow trout (Oncorhynchus mykiss, Walbaum) farms in the province of Mersin, Turkey. These strains were identified by conventional microbiological techniques and by using an ID32GN system. Of these isolates 20 (91.3%) were identified as Aeromonas hydrophila and 2 (8.7%) as Aeromonas sobria. While 8 of the A. hydrophila strains were isolated from water samples, 12 isolates were from fish samples. Whereas A. hydrophila strains were found in all farms, A. sobria was detected in only two farms. Genetic diversity by arbitrarily primed polymerase chain reaction (AP-PCR) and antimicrobial sensitivity tests were carried out on eight A. hydrophila isolates obtained from water samples, and isolates from seven A. hydrophila and one A. sobria from fish samples. The AP-PCR band patterns of motile aeromonads demonstrated weak similarity to the A. hydrophila reference strain ATCC 7966. Genetic Heterogeneity and Antibiotic Susceptibility of Vibrio alginolyticus Strains Isolated from Horse-Mackerel (Trachurus trachurus L., 1758)

Abstract: This study was performed to investigate the presence of halophylic Vibrio spp. in horse-mackerel (Trachurus trachurus L., 1758). In total, 80 fish samples were collected from fish markets and supermarkets between 04.04.2006 and 16.05.2006 in the city of Mersin. All fish samples were microbiologically tested for halophylic Vibrio spp. Nine (11.25%) Vibrio spp. strains were isolated and identified as V. alginolyticus. Molecular epidemiological discrimination and antimicrobial susceptibility patterns of V. alginolyticus strains were also analysed by arbitrarily primed polymerase chain reaction (AP-PCR) and Kirby-Bauer disc diffusion, respectively. Genotypic heterogeneity of these 9 isolates discriminated by AP-PCR determined that while only 3 isolates were from the same genotypic source the other 6 isolates were from different sources. All isolates were resistant to ampicillin, cephalothin, and streptomycin, and susceptible to levofloxacin, ofloxacin, cefotaxime, nalidixic acid, tetracycline, piperacillin-tazobactam, chloramphenicol, imipenem, and aztreonam. Phenotypic and genetic homogeneity of Yersinia ruckeri strains isolated from farmed rainbow trout, Oncorhynchus mykiss (Walbaum), in Mersin Province, Turkey

Twenty-four Yersinia ruckeri strains isolated from farmed rainbow trout, Oncorhynchus mykiss (Walbaum), in Mersin Province (Turkey) were investigated for phenotypic and genetic homogeneity by conventional microbiological techniques, the ID32GN system and arbitrarily primed polymerase chain reaction (AP-PCR). All isolates displayed phenotypic and genetic homogeneity by these techniques.
