Genomewide association of male reproductive traits in Aksaray Malakli dogs

Tuncer, Pürhan Barbaros | Büyükleblebici Serhat

Male reproductive parameters are often used for the functional examination and evaluation of predicted genetic values for future aspects. However, these traits are relatively reliable until the measurable effects are expressed on desired traits. Therefore, we aimed to associate the single nucleotide polymorphism (SNP) geno type of the investigated characteristics and reproductive loci. A total of 46 male dogs aredividedintothreeagegroups(I≤3years,n = 19; II 4–6 years; n = 19, and III ≥7years,n = 8). The testis, scrotum and body weight, libido sexualis and ejaculation time for each fraction were monitored as functional traits, while the pH, fractional semen volume, motility, concentration, and abnormal and dead spermatozoa rate were recorded as spermatological traits. The A...

Influence of Various Antioxidants on Microscopic-Oxidative Stress Indicators and Fertilizing Ability of Frozen-Thawed Bull Semen

Tuncer, Pürhan Barbaros

Cryopreservation is associated with the production of reactive oxygen substances (ROS), which lead to lipid peroxidation of sperm membranes, resulting in a loss of sperm motility, viability and fertility. The aim of this study was to determine effects of the antioxidants of oxidized glutathione (GSSG), reduced glutathione (GSH) and bovine serum albumin (BSA) on standard semen indicators (motility, acrosome and total abnormalities, HOST), endogenous antioxidant enzyme activities and fertilizing ability of frozen-thawed bull semen. Eighteen ejaculates from each of 3 Holstein bulls were collected using an artificial vagina and 9 replicates of the ejaculates were diluted with a Bioxcell®-based extender supplemented with antioxidants, including BSA (5 mg/ml), GSH (2 mM), GGSG (2 mM), and an ext...

Effects of anti-oxidant additives on microscopic and oxidative parameters of Angora goat semen following the freeze–thawing process

Tuncer, Pürhan Barbaros

The anti-oxidant system of reduced glutathione (GSH), glutathione peroxidase (GSH-PX), catalase (CAT), and superoxide dismutase (SOD) has been described as a defense functioning mechanism against lipid peroxidation (LPO) in semen, and is important in maintaining sperm motility and viability. This anti-oxidant capacity of sperm cells may be insufficient in preventing LPOduring the freeze–thawing process. The aim of this study was thus to determine the influence of varying doses of anti-oxidant additives on standard semen parameters, lipid peroxidation and anti-oxidant activities after the freeze–thawing of goat semen. Ejaculate samples (artificial vagina) obtained from 4 mature Angora goats were evaluated and pooled at 37◦C. The semen samples diluted with a Tris-based extender, contai...

Use of Infrared Thermography during Ejaculation Process and Its Link with Semen Quality and Freezability in Dogs

Tuncer, Pürhan Barbaros | Büyükleblebici Serhat

Simple Summary: Scientific attention to infrared technology has grown over the last decade. Remote and non-invasive monitoring techniques are of great importance in discovering ejaculation response and future trends because of their role in vascular flux regulation. However, detailed information about its use in andrology has yet to be fully explained. Therefore, we aimed to reveal information about the amount of sperm to be obtained by observing stress levels with non-invasive eye temperature measurement, and the relationship between various reproductive temperature patterns and parameters of the animal’s various physiological conditions such as age, body condition, total ejaculation time and testicular volume.

Comparison of the effects of glutamine and an amino acid solution on post-thawed ram sperm parameters, lipid peroxidation and anti-oxidant activities

Tuncer, Pürhan Barbaros

Ram semen contains sufficient quantities of superoxide dismutase (SOD) and much lower concentrations of glutathione peroxidase (GSH-PX) and catalase (CAT) to prevent oxidative damage. The anti-oxidant capacity of the sperm cell is limited, due to a small cytoplasmic component, which contains these anti-oxidants to scavenge the oxidants. However ,the concentration of these anti-oxidants may decrease considerably by the dilution of the semen. The aim of the present work was to study the effect of two anti-oxidants, namely ,glutamine and an amino acid solution (BME) in a Tris-based extender on ram sperm parameters, lipid peroxidation and anti-oxidant capacity after the cryopreservation/thawing process. Ejaculates collected from 4 Akkaraman rams were evaluated and pooled at 37◦C. Semen samples ...
