Antioxidative effects of cysteamine, hyaluronan and fetuin on post-thaw semen quality, DNA integrity and oxidative stress parameters in the Brown Swiss bull

Tuncer, P. B. | Buyukleblebici S.

The aim of this study was to compare the effectiveness of antioxidants including cysteamine (2.5, 7.5 mM), hyaluronan (0.25, 1 mg ml 1) and fetuin (5,10 mg ml 1) in the freezing of Brown Swiss bull semen. The best percentagesof CASA motilities were achieved with 10 mg ml 1 of fetuin and 2.5 mM of cysteamine. For sperm morphology, 10 mg ml 1 of fetuin and 2.5 mM of cysteamine had better protective effects (P < 0.001). The results of hypo-osmotic swelling test showed that the percentage values of membrane integrity in all the groups, excluding that supplemented with 5 mg ml 1 of fetuin, were higher than those of the control group (P < 0.001). Results obtained for the DNA damage of sperm cells demonstrated that 0.25 mg ml 1 of hyaluronan, and 2.5 and 7.5 mM of cysteamine led to lower rates o...

Red pine (Pinus brutia Ten) bark tree extract preserves sperm quality by reducing oxidative stress and preventing chromatin damage

Tuncer,, Pürhan Barbaros

This study aimed to investigate the effectiveness of using red pine bark tree extract (P;Pinus brutia Ten) as a TRIS extender in an attempt to prevent oxidative stress in bull spermatozoa after freezing. Semen specimens were obtained from Simmental bulls via an artificial vagina and pooled. They were separated into five specimens and diluted with Tris extender consisting of P (200,100,50 and 25µg/ml) and P free (control ; C) up to a final concentration of 16×106 per straw. All specimens were equilibrated for a period of 4h rata temperature of 4°C ,following which they were filledin 0.25-ml French straws and frozen. Addition of P resulted in favourable tai llength incomparison with C (p.05). Inconclusion, it has been shown that although P added to a Tris extender does not have a pos...

Effects of curcumin and dithioerythritol on frozen-thawed bovine semen

Tuncer, P. B.

The aim of this study was to determine the effects of curcumin and dithio erythritol added into bull semen extender on sperm parameters, lipid peroxida tion, total glutathione and antioxidant potential levels of bull spermatozoa following the freeze/thawing process. Twenty-seven ejaculates obtained from three bulls were included in the study. Each ejaculate that was splitted into five equal groups and diluted in a Tris-based extender containing curcumin (0.5 and 2 mm), dithioerythritol (0.5 and 2 mm) and no additive (control) was cooled to 5 C and frozen in 0.25-ml French straws. The extender supple mented with 0.5 mm dose of curcumin led to lower percentage of total abnor mality (20.40 ± 2.36%) when compared to the control (30.60 ± 1.47%, P < 0.05). Curcumin and dithioerythritol...

Supplementation of quercetin for advanced DNA integrity in bull semen cryopreservation

Tuncer, B. P.

The aim of this study was to identify the effects of adding quercetin (Q) to Tris ex tender in order to identify levels of oxidative stress in bull sperm after freeze thawing. Ejaculates were collected via artificial vagina from Holstein bulls. Semen was divided into five tools and diluted to a final concentration of 15 × 106 spermatozoa/ml with the Tris extender containing Q (25, 50, 100 and 200 μg/ml) and no additive (control; C). All examples were equilibrated at 4°C during 4 hr then were loaded into 0.25- ml straws and frozen using a controlled rate. Sperm motility and motility characteristics were determined using the computer- assisted semen analyser. Sperm membrane in tegrity was assessed using the hypoosmotic swelling test. Sperm chromatin integrity was investigated...
